Carboxypeptidase B (CPB) (EC3.4.17.2) is an exocrine protease, belonging to the family of metallocarboxypeptidase A and carboxypeptidase B (CPA / CPB). Each molecule contains a Zn atom, which can selectively hydrolyze arginine and lysine at the carboxyl end of protein or peptide. Carboxypeptidase B preferentially cleaves basic amino acids, but CPB can also cleave some non basic amino acids. The molecular weight of CPB is 35kD, the isoelectric point is 6.0, and the optimal pH is 7-9. The activity of CPB is competitively inhibited by lysine or arginine, and the activity of CPB is inhibited by metal ion chelating agents such as EDTA. The application of carboxypeptidase B in scientific research is mainly based on its cutting characteristics. It can be used to sequence proteins and peptides and to diagnose pancreatitis in medicine. In addition, in the production of recombinant insulin, the production of active insulin also depends on the cutting of carboxypeptidase B.
